ABSTRACT

The nucleic acid detection is an effective way for the prevention and control of COVID-19. PCR amplification is an important process in the nucleic acid detection. At present, PCR amplification has the problem of low heating/cooling rates, and poor temperature uniformity. This paper proposes a microchannel temperature control device for the nucleic acid detection. Five groups of parallel serpentine channels are used to increase the cooling rate of the PCR amplification. A gradual thermal conductivity design is applied to the reaction module to increase the temperature uniformity. The experimental results show that the best temperature uniformity is obtained when the materials of the inner and outer layers of the reaction module are copper and aluminum alloys, respectively. The limit and average heating/cooling rate are 7.2, 6.12, 5.52 and 5.28 °C/s, respectively, when the input power of the thermoelectric cooler is 11.07 W/cm2, the temperature and flow rate of the cooling water are 15℃ and 700 ml/min, and the thermal conductivity of the thermal grease is 6 W/(m·K). Compared with the commercial fan-fin cooling method, the limit and average heating/cooling rates are increased by 38.02%, 80.82%, 86.49% and 208.77%, respectively, with the help of microchannel cooling method. © 2023 Elsevier Ltd

ABSTRACT

This paper focused on the impact of lifestyle changes in response to the novel coronavirus infection (COVID-19) on the electricity demand of 1398 detached houses during Apr. 2020–Sep. 2020. The rate of increase in power consumption excluding space cooling was the largest in April with an average of 19% for all households. The reason of this increase was analyzed with questionnaire survey. 87 households with room air conditioners consumed more space cooling power in August than usual at an increase of 23%. Power consumption of the centralized air conditioner was less affected than that of the room air conditioner.

ABSTRACT

The open carrier system (OC) is used for vitrification due to its high efficiency in preserving female fertility, but concerns remain that it bears possible risks of cross-contamination. Closed carrier systems (CC) could be an alternative to the OC to increase safety. However, the viability and developmental competence of vitrified/warmed (VW) oocytes using the CC were significantly lower than with OC. We aimed to improve the efficiency of the CC. Metaphase II oocytes were collected from mice after superovulation and subjected to in vitro fertilization after vitrification/warming. Increasing the cooling/warming rate and exposure time to cryoprotectants as key parameters for the CC effectively improved the survival rate and developmental competence of VW oocytes. When all the conditions that improved the outcomes were applied to the conventional CC, hereafter named the modified vitrification/warming procedure using CC (mVW-CC), the viability and developmental competence of VW oocytes were significantly improved as compared to those of VW oocytes in the CC. Furthermore, mVW-CC increased the spindle normality of VW oocytes, as well as the cell number of blastocysts developed from VW oocytes. Collectively, our mVW-CC optimized for mouse oocytes can be utilized for humans without concerns regarding possible cross-contamination during vitrification in the future.
